- Abstract
The long-term vision of this startup research programme is to develop pipelines for multiscale computational modelling based on bottom-up principles that will enable mechanistic interpretation for the changes in cell and tissue excitability induced by high-intensity pulsed electric field (PEF). The first step is to follow up on our recent findings obtained with bioengineered S-HEK cells as minimal excitable systems and establish a theoretical framework that will enable us to model their response to high-intensity PEF. By capitalizing on recent progress in mathematical modelling of cell and tissue excitability, together with our extensive experience in multiscale modelling of cell membrane electroporation, we will establish the foundation for development of computational tools that can be applied to support the advancements in clinical treatments utilizing PEF in excitable tissue. These treatments include ablation of arrhythmogenic cardiac tissue and delivery of nucleic acids to muscle tissue for gene therapy or DNA vaccination.

The project consists of two work packages (WPs), WP1 is devoted to the development of computational models of excitable cells and WP2 to the experimental work needed to validate the models.
